" No. The DOT is independent of the hit. Cold Snap/Vortex Hit dealing some damage and do DOT as well. The DOT itself isn't a hit so it cannot penetrate. Penetration isn't a buff it is a dmg modifier for hits, so if it is a hit it will increase the dmg (dealt by itself) otherwise it do not work. Meanwhile, you can try use Frost Bomb it reduce the monster resistance (like a curse, it's a debuff) increasing cold damage dealt by hits or DOTs. Note that Frost Bomb debuff increase the damage in the same way a cold penetration gem would do.
