Some quick noobish questions...

I didn't see a noob forum, so hopefully this is the right pace for this.

1. Why is it sometimes when I return to the same map I just left, the map is completely reset, yet other times, the map stays the way I left it?

2. Why is it i sometimes lose quests that I have accepted? Such as I had one quest where i was supposed to go into some warriors tomb and return someone, . I went back o town to clear my inventory up because i was full and both the quest and the dude who gave it to me seemed to vanish from that map.

3. Should I be leveling up skills that I find even if I don't intend to use them, or just leave them at level 1?

4. Are all skills you can get able to found from drops/trading, or are there any that can only be obtained through quest rewards?

This game is instanced. When you enter a zone for the first time, you make a new instance of it. Once you leave that zone, the instance starts to expire. Instances expire in 15 minutes for highly connected zones or 8 minutes for side areas. Some instances in the endgame last longer, but don't worry about that for now. So basically, you can't leave a zone for more than a few minutes or it will reset.

(While we're on the subject, you can bring up the instance manager for a zone by ctrl-clicking the zone entrance or waypoint. There you can choose which instance to enter, or create a new instance of the zone. It's quite handy.)

Those are master missions. There are a number of masters who randomly pop up in the world and give you randomly generated missions. Completing them gives you reputation with the master, which makes them offer you all sorts of helpful services. See here. If you let the instance reset, they will disappear with it as their appearance is entirely random.

There is some small value to levelling up gems just for the sake of it, but it's not particularly important and will only come to fruition after hundreds of hours of play. Stick to what you plan to use for now.

No gem is quest-reward specific. Some can only be obtained via drop (or trade, of course), however.

(As a side note, there is no such thing as an untradeable item in this game, except for quest items, so everything can be obtained via trading.)

1. This game is 100% online only. I believe it's kind of expensive to create and maintain map files for all the thousands (millions?) of players out there but also allow these players to refresh maps (to farm for exp, for example), so the simplest, easiest, most sensible solution is using instances. Instances usually last 8 to 15 minutes, but some special instances live for 30 minutes. If they are empty for longer than their lifespan, they are lost to the void forever.

2. See 1, I guess? Don't worry too much about Master missions though - you'll run into that master again and have the chance to get more exp with him.

3. It's your choice. Max level gems have more value than level 1 gems, but it takes a ton of exp to go from 1 to 20 anyways. It may also be harder to sell lv. 2-19 gems than 1 or 20.

4. It's kind of the opposite, actually. All gems in the game can be traded. Nearly all gems can be dropped. Most gems can be obtained through quest rewards. See this page for drop-only gems: http://pathofexile.gamepedia.com/Drop_Only_Gems

See the link on that page for quest gems. All quest gems can also drop.

Quest gems + drop only gems is almost 100% of gems in the game, but there is at least one other that I know of - Block Chance Reduction on this page: http://pathofexile.gamepedia.com/Vendor_recipe_system which can only be gotten from a vendor recipe.
