Does Patient Reaper (trickster) work with..

Does Patient Reaper ascendancy work with killing monsters who are poisoned IF you get the last hit with something other then the poison? IE a melee kill.

Yes. It says enemies have to be affected by DoT, not killed by DoT.
Note that Poison only occurs after a Hit, and thus after the kill - a one-shot won't apply Poison to trigger Patient Reaper. | |

I was thinking of switching from Assassin to Trickster to be more defensive to try to level faster past 95. So IF I'm not 1 shotting everything and using a voidheart it would work?

It would trigger, yes.
CantSeeGround, right? Patient Reaper kinda blows chunks for that particular character. You have instant Leech (no effect), no Regen (no effect), and are not built for ES Recharge (pretty meh?). But, that's up to you. Shade Form's real sweet though, a nice defensive package. | |
