Being a new player to the game I would like to know if there is a way to keep a skill active throughout a fight without having to always press the corresponding letter key. In other words can you lock it in?

You can hold the corresponding key and the skill will keep going. Use the "attack in place button" while doing that so your character does not move due to autotargeting(think the default key of that is shift).
