You need to complete most quest to progress your character but some quests are optional. There is a lot of gear so it is not an easy question. Some pieces are good for some builds. It is good to know what stats you need on gear. For a new player probably life and resistances for armor pieces and damage/speed on a weapon. Builds are usually defined by main skill, some mechanic that they are taking advantage of, and character class. For example 'flameblast prolif templar', would be a templar using flameblast and focusing on proliferating burn damage.
