"Unique" items are not one-per-league, they are just the highest rarity of items. They are set apart from magic and rare items by having set mods on them, rather than random mods - Every Merviel's tears will have the "can't die while healing" stat.

Will there be any variability for the set stats on a given unique item similar to legendary items in Diablo 2? For example, if a weapon has ~100% enhanced damage, could it be possible to find another one with 95% or 105%?
Yes, we can control that when deciding the mods. Some will be variable.

Uniques are rare and rares are unique.
I had never looked at it that way, but it's quite amusing that that's almost* true
*it's technically possible to get two identical rares, just REALLY unlikely.
